Як правило, обумовлена вона генетичними причинами, що призводять до синтезу функціонально неповноцінних білків. Перша успішна спроба подолання подібного дефіциту за рахунок введення відповідного препарату пов’язана з відкриттям та впровадженням в медичну практику інсуліну – невеликого пептидного гормону, що регулює цілу низку процесів (рис. 1). Досить швидко з’ясувалось, що препарати донорного чи тваринного походження не здатні задовольнити постійно зростаючий попит як на цей препарат, так і на цілу низку інших білків для замісної терапіїThe functional lack of certain regulatory proteins is the molecular basis of numerous chronic systemic diseases. The possibility of the compensating of this deficiency by corresponding proteins of animals, donors, and transgenic origin made it possible to treat effectively the group of diseases, which were previously incurable. At the same time, the introduction of such preparation led to the appearance of the wide group of complications, caused by the development of immune response both to the injected drugs and to the own functionally deficient proteins. This work is devoted to the consideration of the causes of the immunogenicity of the protein preparations on the basis of modern ideas about the features of proteins’ structure, the mechanisms and consequences of their violation. Particular attention is paid to the spontaneous formation of nano-scaled aggregates as to the key cause to the immunogenicity of the corresponding preparations.
