Анотація: To synthesize a series of mono- and bis-derivatives of spiro-2-oxindole[3,3’]pyrrole and study the antioxidant and anti-inflammatory activity of the compounds obtained
Current research proved that the preparatory method, namely, the three-component cascade transformation of isatin, α-amino acids and dipolarophils based on bis-maleimides is effective for synthesis of spiro-2-oxindole[3,3']pyrrole mono- and bis-derivatives. Series of hexamethylene- and ethylene-N,N'-bis(spiroindole-3,3'-pyrrolo[3,4-c]pyrrole-4'-nitroso-2a,5a'-dihydro-2,2',6'(1H,1'H,5'H)-trione) symmetric derivatives were obtained. The structure of the obtained compounds was confirmed. Investigation of the antioxidant properties of the synthesized compounds revealed the most active substances: 1'-(hexamethylene-N-maleimido)-5'-benzyl-2a',5a'-dihydro-1'H-spiroindole-3,3'-pyrrolo[3,4-c]pyrrole-2,2',6'(1H,3'H,5'H)-trione 1; ethylene-N,N'-bis(spiroindole-3,3'-pyrrolo[3,4-c]pyrrole-4'-nitroso-5'-methyl-2a,5a'-dihydro-2,2',6'(1H,1'H,5'H)-trione) 6; hexamethylene-N,N'-bis(spiroindole-3,3'-pyrrolo[3,4-c]pyrrole-4'-nitroso-5'-benzyl-2a,5a'-dihydro-2,2',6(1H,1'H,5'H)-trione) 7. Compound 6 also showed anti-inflammatory properties on the reference drug level

Анотація: Carbohydrates, in particular monosaccharides, are necessary for the normal functioning of the human body. Therefore, the search for plant sources of available carbohydrates, such as glucose, fructose, arabinose, is relevant today. Aim. To determine the qualitative composition and the quantitative content of monosaccharides of hawthorn fruits. Materials and methods. Extraction of polysaccharides from the raw material was carried out with purified water in the ratio of the raw material – extractant of 1 : 10 and further treatment of the filtrate with 96 % ethyl alcohol. The preliminary identification of sugars was performed by paper chromatography (PC) after acid hydrolysis. The qualitative composition and the quantitative content of sugars were determined by the gas chromatography-mass spectrometry method on an Agilent 6890N/5973 inert device (Agilent Technologies, USA). The total sugars were isolated by acid hydrolysis using 2 M trifluoroacetic acid. For extraction of free sugars the mixture of methyl alcohol solution and the internal standard (the solution of sorbitol) was added to the powdered raw material. Results and discussion. As a result of chromatography (PC) with the reliable samples glucose and fructose were identified in the raw material. By the chromatography-mass spectrometry study the presence of 5 free sugars was detected, among them glucose (2.02 mg/g), fructose (2.21 mg/g) and sucrose (0.23 mg/g) were determined. After acidic hydrolysis arabinose (1.82 mg/g), xylose (3.88 mg/g), mannose (4.25 mg/g), glucose (5.57 mg/g) and galactose (1.31 mg/g) were identified. Conclusions. The composition of sugars in hawthorn fruit has been found. The highest content has been determined for fructose, glucose and mannose

Анотація: To study the qualitative composition and the quantitative content of phenolic compounds of the liquid extract obtained from cleavers (Galium aparine L.) herb. Materials and methods. The extract of cleavers herb was obtained by triple extraction of the raw material with 60% ethanol when heating, followed by the concentration of the combined extracts to the ratio of the raw material: finished product of 1:1. The content of extractives was determined by the gravimetric method. Phenolic compounds were studied by the methods of high-performance liquid chromatography (HPLC) and UV-spectrophotometry. Results. In the chromatographic study of the extract from cleavers herb 11 compounds of phenolic nature were reliably identified, including 5 hydroxycinnamic acids – caffeic, сhlorogenic, neoсhlorogenic, 3,5- and 4,5-dicaffeoylquinic acids; and 6 flavonoids – catechin, quercetin, quercitrin, isoquercitrin, rutin and hyperoside. Rutin (1.89 mg/ml) and сhlorogenic acid (0.78 mg/ml) were predominant compounds. In the spectrophotometric determination of total content of phenolic compounds it was found that the amount for hydroxycinnamic acids calculated with reference to сhlorogenic acid was 1.9 %, for flavonoids calculated with reference to rutin – 0.25 %, for polyphenolic compounds calculated with reference to gallic acid – 1.35 %. The dry residue was 24.63 %. Conclusions. The significant content of extractive substances, flavonoids and у hydroxycinnamic acids with predominance of rutin and сhlorogenic acid in alcohol-water extract from cleavers herb indicate the feasibility of its further pharmacological study

Анотація: To study the antimicrobial action and the quantitative content of phenolic compounds of the alcoholic extract from the cake of the red raspberry fruit (Rubus idaeus). Materials and methods. The object of the study was the extract obtained from the fruit of Rubus idaeus. The content of phenolic compounds was determined by the spectrophotometric method. The study of the antibacterial activity of the extracts was carried out by the agar diffusion method. Results. It has been found that the content of hydroxycinnamic acid derivatives is 2.21 ± 0.02%, flavonoids – 0.82 ± 0.01%, and the amount of phenolic compounds is 4.52 ± 0.02% in the extract of the red raspberry fruit. The results of the studies conducted have shown that the test sample of the extract from the raspberry fruit has a wide range of the antimicrobial activity, including the antibacterial and antifungal activity. Conclusions. By studying the antimicrobial activity the possibility of creating a new antimicrobial drug based on the raspberry fruit has been proven. The content of phenolic compounds in the extract from the cake of the red raspberry fruit has been studied

Анотація: The properties of pastes depend on a number of factors, the most important of them are the concentration of the solid phase and the nature of the dispersion medium. Aim. To study the effect of the amounts of active substances and excipients on the pharmacotechnological properties of a natural zeolite paste (clinoptilolite) and to optimize the composition of the finished dosage form. Materials and methods. The objects of the study were model compositions of pastes containing the active substance – natural zeolite powder (clinoptilolite) in the concentrations of 20-30 %, a thickener – silicon dioxide in the concentrations of 6.0-8.0 %, and a plasticizer – glycerol in the concentrations of 6.0-8.0 %. Samples of pastes also contained flavoring agents – sorbitol and apple flavoring of 3 % each, and 0.02 % of a preservative – sorbic acid. To select the pharmaceutical factors significantly affecting the study object the fractional factorial experiment 23 was used with equal duplication of experiments. Three quantitative factors were studied. The responses were consistency of the paste, effective viscosity at the shear rate Dr = 5.0 s-1, the thixotropic coefficient and the adsorption activity in relation to methylene blue. The relationship between factors and responses was expressed and interpreted using the regression equations. Results and discussion. It has been determined that the samples studied belong to the thixotropically structured pseudoplastic systems with the structure of the coagulation type. It has been proven that natural zeolite (clinoptilolite) together with silicon dioxide has a structuring effect on the disperse system and the adsorption activity of the paste as a whole; the following composition is optimal (g): natural zeolite (clinoptilolite) 25.0; silicon dioxide 7.0; glycerol 5.0; sorbic acid 0.02; sorbitol 3.0; apple flavor 3.0; purified water 56.98. Conclusions. The studies on the choice of the quantitative content of excipients for obtaining the natural zeolite paste have been conducted; the optimal composition of active substances and excipients has been proposed.

Анотація: To assess the state of pharmaceutical provision for patients with cardiovascular diseases (CVD) using ABC, VEN and complex ABC/VEN analyses. Materials and methods. The study was performed according to the data of the medical records of outpatients who were registered in the Public Health Institution (PSI) “Kharkiv City Clinical Hospital No. 27”. The methods of ABC, VEN and complex ABC/VEN analysis were used. Results and discussion. The results of the study using the ABC-analysis method allowed us to determine that the largest number of drugs was in the group C, which included 25 drugs, or 51.02 % of the total number of prescriptions. During the VEN analysis it was found that the vast majority of drugs (41 drugs) belong to the group V – vital essential medicines. The complex ABC/VEN analysis showed that drugs with the A/V status (the most costly and vital essential medicines) had the highest percentage (32.20 %) of the total consumption. It should be separately noted that the A/N group (the most costly and nonessential medicines) took the second position by percentage after the A/V group. A high percentage (31.05 %) in the A/N group is explained by the fact that the group includes the drug Magnikor, which has the highest frequency of prescription although this drug is not included in the National List of Essential Medicines (NLEM) and in the “Available medicines” program. Conclusions. The results of the studies of the state of pharmaceutical care for patients with CVD make it possible to draw attention to the urgency of the problems of drug availability to the population, namely expanding the list of medicines that are reimbursed in the framework of the government programs, in particular by including the drug Magnikor in the “Available medicines” program

Анотація: Diabetes mellitus (DM) is a national health priority in the world and in Ukraine too. Aim. To substantiate the choice of regimens for combined therapy of type 2 DM (T2DM). Materials and methods. The research materials were retrospective analysis of 1792 medical histories of the inpatients with T2DM. These patients were treated in the endocrinology clinics of the Podolsky region of Ukraine. The methods of frequency analysis; ATC/DDD-methodology; cost-effectiveness analysis were used. The cost of a defined daily dose (DDD) of drugs was studied in UAH according to the data of the average retail price of drugs in Ukraine as of May 2017. Results and discussion. The frequency analysis showed that the monotherapy with metformin, glimepiride and gliclazide was used in 25 % of cases, and combined therapy with metformin + glimepiride, metformin + gliclazide and metformin + glibenclamide was used in 66 % of cases when treating T2DM. It was found that inpatients with metformin + glibenclamide regimen were significantly older, with the longer duration of T2DM, with the highest body mass index and highest levels of fasting plasma glucose (FPG). When comparing inpatients with metformin + gliclazide regimen and metformin + glimepiride regimen a significant difference between the duration of the disease and the level of FPG on admission was determined, no other significant differences were found by other parameters analyzed. The ATC/DDDmethodology showed that metformin + glimepiride regimen had the minimum cost of DDD, and metformin + gliclazide regimen had the maximum cost of DDD. The cost-effectiveness analysis showed that the costs-efficiency ratio (CER) for metformin + glibenclamide regimen varies from 560.10 to 2138.49 UAH, for metformin + glimepiride – from 821.07 to 2300.20 UAH, for metformin + gliclazide – from 798.65 to 2128.60 UAH in the context of minimal and maximal prices of generics. Conclusions. The frequency analysis showed that the combined therapy of T2DM was used in 66 % of cases. It was found that patients with metformin + glibenclamide regimen were significantly older, they had a longer duration of T2DM, the highest BMI and the highest levels of FPG. The regimen of the combined therapy with metformin + glibenclamide has the cost-effective advantages compared to other treatment regimens

Анотація: The aim is to investigate the specific wound-healing effect of a new medicinal form – ointment with groated leaves of Black Alder on the model of a plane wound. Materials and methods. The wound healing effect of the new ointment with groated leaves of Black Alder was carried out on the model of a full-layer planar skin cut wound in rats with planimetric and morphological parameters in comparison with the preparation «Wundehyl». Results and discussion. It is established that the wound healing activity of the ointment with groated leaves of Black Alder was 1.84-4.76 times higher than the ointment «Wundehyl» in the rate of wound healing on the 9-15th day of application. Morphological analysis showed intensive epithelialization and development of granulation tissue during treatment with a new ointment. Conclusions. A new ointment with groated leaves of Black Alder showed the best wound healing effect on the control group and exceeded the reference preparation by 3.33 times, accelerating the healing of wounds for 3-4 days. The test dosage form can be considered a promising treatment for wounds

Анотація: To study the nephroprotective effect of sulfur-containing amino acids (ademetionine, taurine and glutathione) on different models of acute kidney injury (AKI) by the integral index of the survival rate of animals. Materials and methods. The experiments were performed on 105 non-linear mature white laboratory rats and 35 mice. The drugs studied – ademetionine (Geptral, Abbott SpA, Italy) in the dose of 20 mg/kg, glutathione (TAD 600, Biomedica Foscama, Italy) in the dose of 30 mg/kg and taurine (Sigma-Aldrich, USA) in the dose of dose 100 mg/kg – were injected intraperitoneally 3-7 days before AKI modeling. Rhabdomyolysis-induced AKI was modeled by the intramuscular injection of 50 % glycerol solution in the dose of 10 ml/kg, toxic AKI – by the injection of gentamicin solution in the dose of 80 mg/kg and the subcutaneous injection of ethylene glycol in the dose of 10 ml/kg. Simulation of renal ischemia was performed under general anesthesia (ethaminal sodium, 40 mg/kg) by clamping of both renal pedicles for a period of 60 min, followed by reperfusion for 24 h. Results and discusion. The experimental results show a significant improvement in the survival of animals under the effect of the drugs of derivatives of sulfur-containing amino acids studied in conditions of various models of AKI. Ademetionine, taurine and glutathione reduced the lethality of animals on models of rhabdomyolytic, gentamicin, ischemia-reperfusion AKI. In conditions of ethylene glycol intoxication the highest nephroprotective activity by the survival criterion was shown by glutathione (57.1 % survival at 48 h after administration of ethylene glycol against 100 % of mortality in the control group (p 0.05). Conclusions.
