Аннотация: The aim of the research was to study the amino acid profile of the phaseolus vulgaris pods and extract prepared of them. Materials and methods. Five samples of raw material of phaseolus vulgaris pods (erect herbaceous bushes varieties with white seeds) were collected in the Ternopil and Volyn regions, dry extract of phaseolus vulgaris pods was obtained according to previously developed technology. The study of amino acid composition of the raw materials of phaseolus vulgaris pods and extract prepared of them was carried out using a thin-layer chromatography (TLC) and a high-performance liquid chromatography (HPLC). Results and discussion. The better separation of amino acids in TLC-research of the raw material of phaseolus vulgaris pods was observed in the system of solvents isopropanol - formic acid - water (40: 2: 10). As a result of the study, aspartic and glutamic acids, glycine, valine, tyrosine and leucine were identified. The amino acid profile of the studied samples of raw material is quite homogeneous in composition: 7 essential amino acids (histidine, threonine, valine, methionine, phenylalanine, isoleucine and leucine) and 8 non-essential amino acids (aspartic and glutamic acids, arginine, serine, glycine, alanine, tyrosine and proline); lysine was found among the bound amino acids in the 4th sample of raw material. Proline predominates in all samples of raw materials among free amino acids. Among the bound amino acids the content of glutamic acid, which is the product of the glutamine hydrolysis, is the highest. The content of glycine, serine and alanine is also high. Among the essential amino acids, leucine, phenylalanine, histidine, threonine, isoleucine, valine were determined in content descending order. The quantitative determination of amino acids in the extract of phaseolus vulgaris pods proved that the content of proline was the highest (12.47 mg / g); the content of some compounds was also high: aspartic (5.41 mg / g) and glutamic (3.41 mg / g) acids, arginine (5.10 mg / g; both in free and bound forms), glycine (1.02 mg / g) and serine (1.04 mg / g). Among the essential amino acids in the extract, the amount of valine (0.80 mg / g), phenylalanine (0.67 mg / g), threonine (0.66 mg / g), leucine (0.63 mg / g) and isoleucine (0.58 mg / g) was a little different. The total content of amino acids in the extract was 3.2%. Conclusions. 1. The amino acid profile of five samples of phaseolus vulgaris pods was studied by the HPLC method. It has been established that the composition is quite homogeneous, and the total content varies within 0.7-1.1%. 2. In the dry extract of phaseolus vulgaris pods the content of 5 essential and 7 non-essential amino acids has been determined. The content of free amino acids in the extract is 0.52%; the total content of free and bound amino acids is 3.2%. 3. When studying the stability and establishing the shelf life of the dry extract of phaseolus vulgaris pods, it is necessary to take into account the presence of free amino acids and protein substances
Методом ВЕРХ вивчено амінокислотні профілі п’яти зразків стулок квасолі звичайної. Встановлено, що склад є достатньо однорідним, а загальний вміст коливається в межах 0,7-1,1 %. 2. У сухому екстракті стулок квасолі визначено вміст 5 незамінних та 7 замінних амінокислот. Вміст вільних амінокислот в екстракті – 0,52 %, сумарний вміст вільних і зв’язаних амінокислот – 3,2 %. 3. При дослідженні стабільності і встановленні терміну придатності сухого екстракту стулок квасолі необхідно враховувати присутність вільних амінокислот і білкових речовин

Аннотация: The effect of penetrating (glycerol, DMSO) and poorly penetrating (PEG-400) cryoprotective agents on labeled amino acids incorporation into de novo synthesized proteins in the cells of mice thymus, lymph nodes, spleen and cell-free rat liver extract was studied. Cryoprotective agents within the range of concentrations which provide a cryoprotective effect were found to inhibit significantly protein synthesis in cell-free systems under investigation. The most effective inhibition was exerted by polymeric cryoprotective agent PEG-400. Cryoprotective agents more efficiently inhibited protein synthesis at a cell level as compared with that in cell-free system that was likely associated with their effect on amino acids transport system. An inhibitory effect of cryoprotective agents on the protein synthesizing apparatus of cells was determined to be Mg2+-dependent and reversible

Аннотация: The concept of “drug likeness” is used when developing drugs for a potential biologically active substance, which must meet some specific criteria, in particular it should be bioavailable. The traditional method of “drug likeness” assessment is verification of compliance with Lipinski’s rule. Aim. To determine the compliance of the “drug likeness” concept for some N-substituted amino acids and identify the quantitative “structure – microbiological activity” relationships. Materials and methods. Using ChemOffice 2016 software the physicochemical parameters determining the bioavailability of some N-substituted amino acids were calculated. Determination of the possible correlations and quantitative ratios of the biological activity data experimentally obtained with the molar refraction (MR) values calculated was conducted using STATISTIKA 8 program. Results and discussion. All compounds studied in their physicochemical properties meet the requirements for new BAS at the stage of testing their biological activity (correspond to Lipinski’s rule). The dependence of the microbiological action of some N-substituted amino acids on MR is maximal for compounds, which MR value is in the range of 2.13-4.53. The growth of all microorganisms was observed for unsubstituted amino acids (MR 2.8). The maximum activity of all compounds studied was observed against gram-positive (B. subtilis and S. aureus), and the less activity was against gram-negative microorganisms (E. coli, P. vulgaris, P. aeruginosa) and fungi (C. albicans). It may be associated with the structural peculiarities of the cellular wall. The MR values calculated correlate satisfactorily with the experimental data of the antimicrobial activity of compounds. Conclusions. Statistically significant values of MR correlation with the values of the antimicrobial activity of some N-substituted amino acids against the microorganisms studied have been determined. It quantitatively confirms the presence of the “structure – activity” relationship in this series of compounds

Аннотация: The aim of the study was to determine the composition of the sclerotium of Pleurotus tuber-regium and to analyze its nutritional potential. Major minerals and micronutrients content of the P. tuber-regium sclerotium were determined. The study has shown fairly high concentrations of potassium and magnesium as major minerals with values of 60.66 ± 4.13 and 41.79 ± 3.14 mg/kg, while manganese and zinc were micronutrients with the highest values of 1.20 ± 0.10 and 0.95 ± 0.07 mg/kg. Glutamic acid and aspartic acid were also observed in high concentrations with values of 11.51 ± 1.01 and 5.52 ± 0.86 mg/kg. The mushroom powder of P. tuber-regium was a source for production of oil, which was analyzed by GC-MS method. Benzenedicarboxylic acid mono-(2-ethylhexyl) ester and benzenedicarboxylic acid butyl-cyclohexyl ester were volatile constituents predominating with percentage total of 78.7 and 5.2, respectively. It is concluded that the presence of mineral elements, amino acids and volatile components observed in this fungus indicated the presence of the nutritional potential in the sclerotia of P. tuber-regium

Аннотация: The study of the reactivity of substances allows optimizing the conditions of their synthesis and creating mathematical models of the “structure-biological activity” relationship that enable to carry out the targeted search of compounds with a specified high level of the biological action. Aim. To study the reactivity of propyl esters of N-[(2-oxoindolin-3-ylidene)-2-oxiacetyl]amino acids. Materials and methods. The reactivity of propyl esters of N-[(2-oxoindolin-3-ylidene)-2-oxiacetyl]amino acids was studied in reversible conditions on the model of acid-base equilibria by the method of potentiometric titration. Results and discussion. The ionization constants of 9 propyl esters of N-[(2-oxoindolin-3-ylidene)-2-oxiacetyl]amino acids indicate that these compounds are weak acids. Elongation of the methylene fragments in the amino acid groups of molecules decreases their acidity. In contrast to the length of the methylene fragments their branching and the presence of the substituents do not affect acidity of the esters studied. In the framework of the principle of linearity of free energies the effect of the length of the methylene chain in the amino acid fragment of the molecules of propyl esters of N-[(2-oxoindolin-3-ylidene)-2-oxiacetyl]amino acids was quantitatively assessed by Hammett equation. Conclusions. It has been found that propyl esters of N-[(2-oxoindolin-3-ylidene)-2-oxiacetyl]amino acids have the functions of weak monobasic acids. Elongation of the polymethylene chain weakens ionization. The low sensitivity of the reaction center to elongation of the methylene chain has been identified
